Clustering of CTLA-4 siRNA and CPG oligodeoxynucleotide encapsulated gold nanoparticles for tumor targeting and immunotherapy |

Small size and highly cationic gold nanoparticles (AuNPs) loaded with CTLA-4 siRNA, CPG ODN, and targeting moieties are assembled to clusters by click chemistry using an alkyne-MMP-2 cleavable peptide as a linker for enhanced tumor targeting and immunotherapy. Branched PEI (bPEI, Mw 10 k) is thiolated by substituting the primary amines. To conjugate azido groups on HS-bPEI for click chemistry, maleimide-PEG-NHS is first reacted with 3-azido-1-propanamine, and then reacted with HS-bPEI. HS-bPEI and HS-bPEI-PEG-N3 are mixed with chlorocauric acid solution and sodium borohydride dissolved in cold water is then rapidly added for synthesis of AuNPs. The AuNPs are highly cationically charged with abundant amine groups and displays azido groups on the surface, and are thus suitable for encapsulation of siRNA, conjugation of targeting antibodies and clustering. The digested AuNPs are expected to be easily internalized by immune cells in tumor site, and facilitate tumor immunotherapy. |
