제목 The enhanced adhesion and proliferation of rabbit BMSCs in RGD-modified MPEG-PCL
초록 Poly(ethylene glycol)-b-poly(-caprolactone) diblock copolymer (MPEG-PCL) was studied as an injectable biomaterial for in situ-forming scaffold. A major limitation of MPEG-PCL for these applications is the lack of cell-binding sites. In this study, to improve the engraftment of cells, Arg-Gly-Asp (RGD) peptide was conjugated to the end group of MEPG-PCL. Rheological measurements showed a gelation temperature of 37℃ for RGD-conjugated MPEG-PCL. Microscopic studies showed that the clusters of BMSCs were encapsulated into RGD-conjugated MPEG-PCL hydrogel, and BMSCs were attached on the networks of hydrogel by focal adhesion. After 3 days of incubation, BMSCs started to spread and the maximum spreadibility was found at 7 days of incubation. Further, the immobilization of RGD to MPEG-PCL promoted BMSCs viability and differentiation.
