제목 Preparation of chemically modified mRNA by in vitro transcription (IVT) for protein expression
초록 Messenger RNA (mRNA) is a genetic material generated by transcription of DNA and these single strand RNA molecules are employed to induce the protein expression of target genes. mRNA can be an alternative genetic material which can be expressed in mammalian cells instead of plasmid DNA (pDNA) in non-viral gene delivery system. It has multiple advantages over pDNA such as 1) less genetic stress on host cells, 2) fast on-set of target gene expression, 3) no nuclear localization is required. In this study, we have prepared chemically modified mRNA by in vitro transcription (IVT) method along with circularization of template DNA and linearization of pDNA template. Various bioreducible polymeric systems have been screened to optimize their mRNA delivery efficiency. Transfection and expression of target genes have been confirmed in mouse embryonic fibroblast (MEF) cells.
