Delivery of messenger RNA (mRNA) has several advantages over delivery of plasmid DNA in production of therapeutic proteins. However, mRNA-based therapies have been hampered by mRNA susceptibility to nuclease degradation and inability to achieve sufficient expression level. Inspired by rolling circle transcription (RCT) method that can generate extremely long RNA strands, we report a novel strategy for synthesizing mRNA nanoparticles (mRNA-NPs) as mRNA delivery module with high potency for the production of desired proteins in cells. mRNA-NPs showed enhanced stability of mRNA in physiological conditions, and successful expression of green fluorescence protein in cells. Moreover, this approach requires only one-step process to synthesize particles with minimized plasmid DNA to produce RNA transcripts via RCT to achieve efficient delivery and high cargo capacity. |
