Microcystis aeruginosa, a photosynthetic cyanobacteria, requires anaerobic condition and the absence of sulfur (S) in the medium to produce hydrogen gas. Therefore, we managed bioreactor operation in a cyclic manner of following two distinct stages; (i) Stage I: growth phase under CO2 and light, (ii) Stage II: H2 production phase in the absence of light, O2 and S. Since cells do not grow and their physiological activity is decreasing, cells that produced H2 for a certain period in Stage II must go back to the growth phase (Stage 1) to accumulate photosynthetic reserves. We immobilized M. aeruginosa cells with agar to achieve a prompt medium replacement and quick change of cultivation condition between Stages I and II. |
