Synthesis of Multivalent (Nitrilotriacetic Acid)-End-Functionalized Polystyrenes and Their Self-Assembly |

High-affinity of nitrilotriacetic acids (NTA) has great potential in the molecular manipulation of His-tagged proteins. And polymer-protein hybrid nanostructures are important for their potential applications in pharmaceuticals and biotechnology. At low concentration applied frequently in biological systems, multivalent NTA derivatives (KD ca. 20 x 10-9 M) are better suited for these applications than mono NTA (KD ca. 10 x 10-6 M) due to the higher binding affinity. In this work, we synthesized (multivalent NTA)-end-functionalized polystyrene using atom transfer radical polymerization (ATRP). Polymerization was carried out to yield well-controlled polystyrenes with multivalent NTA chain end. Then we also studied a simple method for the construction and control of polymer-protein hybrid spherical aggregates in situ from nickel complexed Ni-NTA-PS and His-tagged GFP through NTA-Ni-His interaction in water-DMF (DMF 4 vol %) at physiological pH. |
