제목 Multi-responsive Protein-polymer Hybrid Hydrogels with Dynamic Property
초록 Genetically engineered polypeptides with stimuli-responsiveness have been of great interest because of accurate control over molecular design, biodegradation, biocompatibility, and biological functions. Their hybrid materials with biocompatible polymers would be interesting for a variety of biomedical applications. We report herein that multi-responsive protein-polymer hybrid hydrogels were formed by covalent crosslinking of multi-responsive fusion proteins having thermally-responsive elastin-based polypeptide, calcium- and ligand-responsive calmodulin, and cysteine domain with poly(ethylene)glycol (PEG) derivatives. The multi-responsive fusion protein was genetically designed, expressed, and purified by inverse transition cycling (ITC), followed by chemical crosslinking with biocompatible PEGs. The dynamic hybrid hydrogel was affected by temperature, calcium-ion, and ligand, which would be useful for functional tissue engineering and drug delivery.
