Label free analysis of multivalent ion binding using bioresponsive hydrogels and SPR |

Metal ions play important regulatory roles in the cellular and subcellular systems. For instance, divalent ions such as magnesium and calcium are known to regulate enzymes for various chemical reactions. Hence, accurate determination of metal ion binding events is required for better understanding of the enzymatic reactions in the biological system. Here, we report a method to distinguish between monovalent and multivalent metal ion binding. With the use of a hydrogel modified SPR chip, we were able to show that in comparison to monovalent ions, multivalent ions in the +2 and +3 oxidation states cause a greater extent of amplification in SPR signals. Hence, our results suggest that the evaluation of the oxidation state of metal ions is possible through SPR analysis. |
