Light Activated Adhesive Hydrogel Originated from Mussel Adhesion Protein for Cornea Wound Repair |

Development of cornea wound healing method is necessary for delayed cornea regeneration and re-epithelialization because injury can cause epithelial defects, corneal trachoma, and ulceration. Currently used suturing cannot perfectly seal the wound and cause scar on tissue and existing surgical glues do not satisfy the practical requirement for successful wound healing due to lack of wet adhesiveness and cytotoxicity. In this research, biocompatible and wet adhesive protein glue using mussel derived recombinant adhesive protein (MAP) was applied on cornea wound repairing in form of light-activated bioadhesive hydrogel by using dityrosine crosslinking. The rapid cross-linkable light-activated MAP hydrogel could maintain the intraocular pressure by seal the wound in short time. During the cornea perforation blocking test using porcine eyes, the applied MAP successfully form hydrogel and maintain formation on cornea perforation after mold was detached. Also the MAP hydrogel showed higher wound closure ability than the commercialized surgical glue due to its wet adhesive and fast crosslinking properties. The cytotoxicities of MAP hydrogel were evaluated by In-vitro human corneal epithelial cells (hCECs) cultivation. Consequently, the cells were well adhered on MAP hydrogel showing fine viability. The platform using MAP-based hydrogel would be the attractive solution for cornea wound surgery material. |
