Response regulator provides resistance and tolerance against abiotic stresses. The effects of RR from extremophiles could be utilized in industries for the development of recombinant strains. In this study, RR homologs were mined from NGS data of Antarctic bacteria. DR1558, one of the RRs from Deinococcus radiodurans, an extremophilic bacterium known as one of the most radiation-resistant organisms, showed advanced tolerance toward H2O2, γ-radiation, heat, osmotic pressure, and acidic stress. We investigated amino acid sequences of RRs related to DR1558 by BLAST and NGS and 3 amino acid sequences were selected as putative homologs. The ORFs were named DRH632, DRH1601, and DRH577. Three amino acid homologs were ligated onto pRadGro; pRadGro_drh632, pRadGro_drh1601, pRadGro_drh577, respectively. These plasmids were transformed into Escherichia coli DH5α and transformant strains were subjected to various abiotic stresses. As a result, E. coli DH5α/pRadGro_drh632 exhibited the highest survival rate in osmotic, acidic, oxidative, and hypothermic stress and E. coli DH5α/pRadGro_drh1601 exhibited the highest tolerance level to osmotic stresses. |
