Coenzyme B12 has an essential role in biological synthesis of chemicals such as 3-HP. Pseudomonas denitrificans is well known as industrial B12 production strains. Herein, we analyzed and characterized cobalamin gene clusters in P. denitrificans, in terms of transcriptional regulation by various B12-riboswitches and their essentiality for B12 production. In vitro characterization of five B12 riboswitch structures identified in P. denitrificans showed that only three riboswitches were transcriptionally regulated. Further, the essentiality of uncharacterized genes to B12 synthesis were also evaluated. Interestingly, only the gene bgpM was found to be essential for B12 biosynthesis. Surprisingly, cobA deletion mutant, an important branching point in B12 biosynthesis pathway could still synthesize coenzyme B12. Based on the results, we will have a better understanding about B12 synthesis to further engineer B12 pathway in P. denitrificans for several industrial applications. |
