Biodegradable polymer nanoparticles were synthesized by the ionic assembly of charged polymers and indocyanine green (ICG) dye. We introduced biocompatible polymers, poly-γ-glutamic acid (PGA) and poly(β-amino ester)s (PBAE). PBAE has good transfection efficiency and promoted degradation in acidic conditions, which was used for effective release of encapsulating materials after cellular uptake. As an encapsulating material, ICG is FDA approved near infra-red dye and has a quenching property in high concentration. In this regard, we focus on the rapid degradation of PBAE in the cells, which helps ICG dyes to make enhanced fluorescence signals due to releasing from the particles. Especially, the particles were modified for targeting breast cancer cell, HER-2 positive SKBR3, by conjugation of HER-2 antibody on their surfaces. Effective cellular uptakes were observed by enhanced fluorescence within cytosol. This could give great potential applications for clinical use in the future. |
