제목 A trilayer structured nanofilm with efficient encapsulation and sustained release of bFGF to extend medium replacement cycle in human iPS culture
초록 Induced pluripotent stem cells (iPSCs) have the potential to generate patient-specific regenerative medicine. Especially, basic fibroblast growth factor (bFGF) is essential signaling molecules to maintain undifferentiated state and pluripotency of iPSCs. However, the bFGF should be daily supplied to iPSCs culture medium due to its instability in physiological condition. So, we established the nanofilm to maintain the bFGF level in iPSCs culture medium. This nanofilm has layer-by-layer assembled multi-trilayer structure, which provides attractive advantages such as enhanced encapsulation efficiency of bFGF and short build-up time compared with conventional multilayer nanofilm. As a result, the release amount of bFGF from the nanofilm was 50.7ng/cm2 during 3 days. Also, the iPSCs were grown in the presence of the nano-film maintained their undifferentiated morphology and expression levels of pluripotency marker proteins despite of tri-daily replacement of growth medium.
