Reduction of Ag-PVP toxicity using biologically-derived materials ; Lung epithelium 2D, 3D cell culture sensitivity |

In this study, we have cultured 3D cells as an in vitro model that provides an environment similar to the in vivo environment of the 2D cell model, and compared the risk assessment differences for 2D and 3D cell nano-toxicity. Experimental results showed that 3D cells grew by forming spheroids in the gel. ICP-MS analysis of the gel revealed that the inner layer had a lower concentration of Ag NPs than the outer layer of the gel. In addition, there was no significant difference between the control and exposed groups in the cell survival analysis, confirming that the 3D cells were less affected by the toxicity of Ag-PVP NPs than the 2D cells. The structure of the spheroids in the gel of the 3D cells cultured in this study can reflect the characteristics of the tissues constituting the layer by density of the cells with high density and can carry out the risk evaluation more reliably than 2D cells It is judged. |
