제목 Preparation and characterization of ovomucin/gelatin hydrogels for tissue engineering.
초록 Ovomucin is a sulfated glycoprotein that comprises 3.5% of the total egg white proteins that is responsible for the gel like property of egg white. This study aims to fabricate hydrogels made of ovomucin and gelatin from porcine and evaluate its potential for tissue engineering. Ovomucin was extracted from fresh egg whites using the two step isoelectric precipitation with 100mM and 500mM NaCl at pH 6.0.  Different blends of ovomucin/gelatin hydrogels were fabricated with 0%, 30%, 50%, 70% and 100% ovomucin then crosslinked with genipin. Scanning electron microscopy (SEM), porosity and pore size distribution analyses were done to visualize the morphology and structure of the samples. Mechanical strength, swelling ability and in vitro biodegradation were also tested on the samples. Mesenchymal stem cells were seeded on the samples to observe the cell proliferation and adhesion. MTT assay was conducted 1, 3 and 7 days to quantify cell proliferation and live/dead cell assay for visualization. Cell adhesion behavior on the hydrogels was observed using confocal laser scanning microscopy and SEM. Ovomucin from hen egg white, stabilized by gelatin to harden at room temperature, is a potential material for tissue engineering.
