Zinc protoporphyrin IX (ZnPPIX) has been attracted colorant for production of nitrate/nitrite free meat. It is the analog of heme, ZnPPIX has Zinc central metal ion Zn2+ instead of Fe2+. However, the production of ZnPPIX has not been reported yet. Metabolically engineered Escherichia coli has previously been reported secretory production of heme. In this study, we repurposed this strain as ZnPPIX producing strain using various culture condition. To produce ZnPPIX and minimize production of heme, the component of culture medium was firstly screened at flask level. Next, we optimized the other factor and culture conditions during fed-batch fermentation process. As a result, ZnPPIX producing strain produced high titer of ZnPPIX in more efficient productivity than heme production during fed-batch fermentation. The optimization strategies employed here is applicable for the high-level production of bio-based production of safer red-colorant for alternative meats.
