Non-covalent method was used for bioconjugation of Ni2+ complexed nitrilotriacetic acid (NTA) end-functionalized polymers with genetically modified six histidine-tagged green fluorescent proteins (His6-GFP). NTA end-functionalized poly[poly(ethyleneglycol) methyl ether methacrylate] (NTA-p(PEGMA)) was prepared using initiators containing tert-butyl protected NTA moiety via atom transfer radical polymerization (ATRP). Well-defined NTA-p(PEGMA) was characterized using 1H NMR, and GPC. Using end-functionalized polymers with amphiphilic structures, stabilization of proteins was attempted due to the formation of micelles in the solution. D-values for the stabilization of protein by conjugation with end-functionalized polymer were determined using fluorescent spectroscopy. The micelles of conjugates were observed using transmission electron microscopy (TEM) observations and dynamic light scattering (DLS). |
