Abstract: Tissue engineering has the possible to improve upon current techniques for intervertebral disc repair. This study approached for best pore to select suitable pore for engineered disc for 4 weeks in vivo. PLGA scaffolds were prepared by solvent casting/salt-leaching with various pores at 90~180 ul , 180~250 ul, 250~355 ul , 355~425 ul , 425~600 ul. Nude mice were sacrificed at 1, 2, 4 weeks and each samples were harvested at each time point after retrival PLGA scaffolds seeding NP cell were implanted into the subcutaneous space in nude mice. To characterize the histologic appeatance, Safranin-O dye, Immunohistoch emistry (Collagen Type II) dye, compression strength measurement of the engineered scaffolds. The previous study confirmed that scaffolds of big pore size is weak for in vitro, we presupose that this study for in vivo have a same result like the previous study.
