To maintain the enzymatic activity of papain for long-term storage, a buffer layer was introduced in papain microcapsules produced by O/W emulsion solvent evaporation method. Biodegradable polycaprolactone (PCL) and amphiphilic polymer (Poloxamer 407) consisting of a triblock copolymer of poly(ethylene oxide) (PEO)-poly(propylene oxide) (PPO)-PEO, were employed as polymer shell and buffer layer, respectively. By incorporating Poloxamer 407 at the interface of polymer wall and bio-active material, the conformational change of papain owing to chemical and physical stress could be prohibited and sustained its bio-activity. The enzymatic activity of papain microcapsules depending on the content of Poloxamer 407 was measured as a function of time utilizing UV-vis spectroscope. Generation and morphology of capsules were monitored by optical microscope (OM) and scanning electron microscope (SEM). The existence of papain in microcapsules was confirmed by using energy dispersive x-ray spectrometry (EDAX). |
