Development of Chemical Tools for Studying a Novel Glycan-dependent Signaling Pathway |

Dynamic O-GlcNAcylation is an important signaling mechanism akin to phosphorylation. Deregulation of this pathway has been linked to insulin resistance and several neurodegeneration. We developed a labeling approach that has now provided a general means for O-GlcNAc proteome. Attachment and detachment of this O-GlcNAc are carried out by two key enzymes which are O-GlcNAc transferase and O-GlcNAcase, respectively.We developed a high-throughput assay with a highly sensitive fluorogenic hexosaminidase substrate for O-GlcNAcase. This substrate was further optimized to give selectivity for O-GlcNAcase over other related hexoaminidases.We also generated novel inhibitors of O-GlcNAcase with excellent selectivity and potency.Such inhibitors can induce non-insulin-dependent diabetes in model systems making them invaluable research tools. |
