Bone regeneration in rat calvaria defect using porous PCL/BCP scaffolds with surface modification |

To make polycaprolactone (PCL) more suitable for bone tissue engineering, PCL in the form of porous scaffolds was first loaded with biphasic calcium phosphate (BCP) and then modified with 1,6-hexamethylenediamine to introduce amino groups on their surface. Collagen was then coated on their surface, and 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide used as the crosslinker agent. The appropriateness of these porous PCL and PCL-BCP scaffolds for the tissue/cell culture was evaluated in vitro with mouse calvaria-derived preosteoblastic cells (MC3T3-E1). The surface modification of PCL and PCL-BCP scaffolds with collagen showed the good ability to support the attachment, proliferation and protein expression, mRNA expression of the pre-osteoblastcells. The in-vivo study was done by implanting the porous PCL, PCL-Collagen, PCL-BCP and PCL-BCP-Collagen to the skull of rats. Newly bone formed in PCL-BCP-Collagen after 6 weeks implantation were observed by hematoxylin&eosin staining. The PCL-BCP-Collagen hold a potential candidate as scaffolds for the Osteoconduction of bone regeneration. |
