The development of biocompatible materials which can be processed into three-dimensional scaffolds and design of appropriate configuration to enable the cellular infiltration and proliferation are one of the major issues in the tissue engineering. In this study, polycaprolactone(PCL)/hydroxyapatite(HA) scaffolds were prepared to increase mechanical properties and degradation for bone tissue engineering. Initially, PCL macromers were synthesized through the reaction of PCL diol(M.W.:530,1250,2000) and PCL triol(M.W.:900) with acryloyl chloride. The PCL/HA scaffolds were prepared by crosslinking of PCL macromer in the presence of HA by UV treatment and freeze drying methods. Mechanical and morphological characteristics as well as degradability of the PCL/HA scaffolds were investigated. Cell proliferation and cytotoxicity of MG63 osteoblast cultured onto the PCL/HA scaffolds were assessed by Alamar blue and LDH assay, respectively. |
