Microalgae have potential as a source of sustainable energy. However, engineering of microalgae to improve photosynthetic efficiency is restricted by the lack of rapid screening method. Here we show a microscale approach using competitive phototaxis of Chlamydomonas reinhardtii, for efficient isolation of strains with improved photosynthetic efficiency. By exploiting competitive phototaxis based on the beneficial relationship between phototaxis and photosynthesis, we could enrich the population of strains with high photosynthetic efficiency by isolating cells showing rapid phototactic responses from the mixture of 104 mutants. Among the strains isolated after screening, 94.6% showed improved the growth over the parental strain up to 1.9-fold, substantial improvement over previous approaches. Our approach provides a powerful means for rapid isolation of strains with high productivity by enabling one-pot screening of large mutant libraries with broad applicability to strain development. |
