Biocompatibility evaluation of Platelet-rich plasma (PRP) with hyaluronic acid/gelatin-BCP hydrogel loading in BCP sponge scaffold for bone regeneration |

Platelet-Rich-Plasma (PRP) has been widely used in clinical bone regeneration and bone wound healing. Bio-ceramics are comparable to the mineral component of natural bone and showed good osteoconductivity. In this study, 10% Biphasic Calcium Phosphate (BCP) mixed hyaluronic acid-gelatin hydrogel with PRP (HGB10/PRP), and these hydrogel was loaded in BCP sponge scaffold (HGB10) to improve mechanical property and biocompatibility. In vitro studies of BCP, HGB10 and HGB10/PRP scaffolds were evaluated by cytotoxicity, cell attachment and proliferation studies using MC3T3-E1 preosteoblast cells. In vivo studies were investigated with autologous PRP (HGB10/PRP) of rats for 2 weeks, 4 weeks and 8 weeks compared with BCP, HGB10 scaffolds. HGB10 scaffold was higher mechanical property than BCP scaffold. In in-vitro studies, cell count, cell proliferation behavior and live cell of HGB10/PRP scaffold were evaluated higher than without PRP scaffold. There results showed that with HGB10/PRP scaffold had biocompatibility and osteo-inductive properties for the treatment of bone defect. |
