Fabrication and evaluation of platelet-rich plasma (PRP) loaded BCP scaffold for bone regeneration |

The aim of this study is to evaluate the regenerative effects of platelet-rich plasma (PRP) combined with porous biphasic calcium phosphate (BCP) scaffold for bone tissue engineering. PRP is nowadays widely used in different clinical surgery, such as orthopedics and healing therapies. PRP is rich in growth factors, such as transforming growth factor (TGF-β), platelet-derived growth factor (PDGF), insulin-like growth factor (IGF), basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF). PRP and platelet-poor plasma (PPP) were isolated from fresh rabbit blood and loaded on the porous BCP scaffold fabricated by sponge replica method. The scaffolds were characterized by scanning electron microscopy (SEM), electron dispersion spectroscopy (EDS), X-ray diffraction (XRD). SEM results showed that mesh-like microstructure was formed from PRP inside inner pores of BCP scaffolds. Compared with PPP-BCP scaffold, the PRP-BCP scaffold exhibited excellent biocompatibility, cell proliferation and cell adhesion behavior resulted in the MTT assay and immunofluorescence analysis using bone marrow messenchymal stem cells (BMSCs). Based on these results, PRP-BCP scaffolds could be considered as potential bone tissue engineering applications. |
