Preparation and Characterization of Polymeric Anticancer Drug |

In this study, we have synthesized doxorubicin (DOX)-conjugated poly(DL-lactide-co-glycolide) and prepared nanoparticles without use of surfactant. DOX contents in nanoparticles were 1.2 ~ 4.75 % (w/w). At transmission electron microscopy observations, nanoparticles showed spherical shapes with size around 100~200 nm. Drug was continuously released from nanoparticles over one month. Especially, overall drug release from nanoparicles prepared with higher M.W. was significantly faster than that of lower M.W. At cytotoxicity test at invitro using CT26 colon carcinoma cell, free DOX showed higher cytotoxicity than DOX-LG. These results indicated that DOX was slowly released from nanoparticles and exposed to tumor cells, and, due to these properties of nanoparticles, DOX-LG might be showed lower cytotoxicity. |
