제목 Intracellular targeting delivery of anticancer drug with AB2 miktoarm polymeric carrier
초록 The delivery of drugs to the intracellular target organelles such as nucleus and mitochondria is a promising and efficient strategy for getting higher therapeutic efficacy with minimizing side effects. We have designed and synthesized a bioreducible AB2 miktoarm polymer that can form self-assembled nanoparticles (NPs) in aqueous solution and has functional groups available for chemical conjugation with an anticancer drug. For targeting the mitochondria which is known as “power house of cells”, triphenylphosphorinum (TPP) molecule was physically or chemically introduced into the doxorubicin (DOX) conjugated AB2 miktoarm NPs. The nanoparticle size (200-250 nm in diameter) of the DOX conjugated AB2 miktoarm polymer was reduced to 100-150 nm after loading of lipophilic TPP. The DOX and TPP contents in NPs was characterized by 1H-NMR and UV Vis. The DOX conjugated NPs with the chemically or physically introduced TPP were evaluated in terms of intracellular targeting and anticancer effect.
