Recently, to increase the accumulation of photosensitizer in tumor cells, liposomes and nanoparticles are used as carriers to deliver photosensitizers. In this study we investigated the effect of Photodynamic Therapy (PDT) with protoporphyrine IX(PpIX) on MKN28 cell line. Thus, we used octaammonium polyhedral oligomeric silsesquioxane (POSS) as carrier, resulting that a number of PpIX can be introduced for the increasement of endocytosis of photosensitizers. Additionally, for the purpose of lower cytotoxicity in dark condition, fatty acids were also conjugated with those nanospheres. POSS-PpIX-fatty acid(PPFA)-conjugated nanospheres were characterized by FT-IR, UV, DLS.Then, in vitro studies their cytotoxicity, reactive oxygen species(ROS) generation, and uptake were investigated. They showed lower cytotoxicity in dark toxicity compared to free PpIX as shown in MTT assay. Therefore, we propose that PPFA-conjugated nanospheres are promising agent for PDT with reduced side-effects. |
