We previously developed protoporphyrine IX(PpIX)-conjugated nanoparticles(NPs) with saturated/unsaturated fatty acids(FAs). In vitro the cytotoxicity of those NPs examined on human gastric adenocarcinoma(MKN28 cell line) was tested by MTT assay. The results showed that a PpIX-conjugated NP with linolenic acid exhibited lower dark toxicity in comparison to a PpIX-conjugated NP with stearic acid, indicating a potential PDT agent for lower side-effects. It allows delivery of selective photodynamic therapy(PDT) to the cancerous tissues, with minimal harm to the adjacent normal tissues. From these results, we decided to further investigate the different effects of saturated/unsaturated FAs on cytotoxicity of the tumor cell. To compare the effect of saturated and unsaturated FAs, we conducted 1)cellular uptake, 2)intracellular reactive oxygen species(ROS) generation, and 3)cell death mechanism experiments, assumed to be involved in the cytotoxicity of fatty acids on tumor cells. |
