There has been much interest in producing natural colorants to replace synthetic colorants, due to the increasing awareness of environmental and health concerns. Majority of natural products are known to be hydrophobic and accumulate within the cell membrane that has limited storage space and also inhibits cell growth. In this work, hydrophobic natural rainbow colorants were produced through application of systems metabolic engineering and membrane engineering strategies. Through further process optimization, the engineered strains were able to produce 322 mg/L of astaxanthin (red), 343 mg/L of β-carotene (orange), 218 mg/L of zeaxanthin (yellow), 1.42 g/L of proviolacein (green), 0.844 g/L of prodeoxyviolacein (blue), 6.19 g/L of violacein (navy), and 11.26 g/L of deoxyviolacein (purple). The membrane engineering strategies reported here are applicable to the microbial production of a wide range of hydrophobic natural products [This work was carried out with the support of "Cooperative Research Program for Agriculture Science and Technology Development (Project No. PJ01550602)" Rural Development Administration, Republic of Korea]. |
