The ferritins (FRTs) is a major iron storage protein that naturally occurs in the human and living organism. Each ferritin is composed 24 subunits which self-assemble to form a cage-like nanostructure. The Ferritin protein is composed two type of subunits H type (heavy) and L-type (light) they are encoded by a separated gene with nonexchangeable function. The Iron Free ferritin is called Apoferritin (AFTN), it has been produced by chemically reducing iron within the ferritin. The resulting apoferritin (AFTN) protein expression from Escherichia coli bacterial host cell while higher efficiency depends on a great copy of the gene. Apoferritin subunits are genetically modified to expression protein G, and 6-His tag protein functionalizes on C terminals site. The modified Apoferritin units can be conjugated with surface functionalized Ni-NTA interaction. Each subunit expresses protein G and 6-His tag surface protein which interacts with Fc region of an antibody. Specially Apoferritin nanoprobe can easily utilize for detection of pathogenic microorganisms. |
