We have studied the self-assembly behavior of N-hydroxysuccinimide (NHS) ester-end-functionalized amphiphilic block copolymer, NHS ester-poly(poly(ethylene glycol)methyl ether methacrylate)-block-poly(methyl methacrylate) (NHS-p(PEGMA-b-MMA)), and protein immobilization on surface of the resultant polymeric micelles. We synthesized NHS-p(PEGMA-b-MMA) by Atom Transfer Radical Polymerization (ATRP) method. To induce self-assembly, water was slowly added to the copolymer/THF solution as selective solvent. After evaporation of THF, the well-defined spherical micelles indicating NHS ester moieties on their surface was generated from NHS-p(PEGMA-b-MMA). Through covalent bonding between lysine of the protein sequence and NHS-ester of polymer, protein-polymer hybrid nanostructure was successfully obtained. |
