Hydrogenotrophicmethanogens can use gaseous substrates like H2 and CO2 in methane production. We successfully have operateda novel hollow-fiber membrane biofilm reactor (Hf-MBfR) for stable methane production from H2 and CO2 undercontinuous operation. H2 and CO2 were diffused into waterthrough the membrane without bubbles formation in theHf-MBfR operatedat pH 4.5~5.5 over 70 days. Focusing on the presence of hydrogenotrophic methanogens,we analyzedthestructureof the microbial community in the reactor. Denaturinggradient gel electrophoresis was done with bacterial and archaeal 16S rDNAprimers. Real-time qPCRwas performed to check the variance inthe methanogens according to the operation time.Finally, the microbial community at a condition with maximum methane productionwere analyzed by pyrosequencing methods. The results showed that acidicoperation of a methane production reactor without any pH adjustment inhibitedacetogenic growth and enriched the hydrogenotrophic methanogens, decreasing thegrowth of acetoclastic methanogens. |
