제목 A novel pluronic grafted HA hydrogel for injectable cell-therapy
초록 A novel pluronic grafted hyaluronic acid (HA) was investigated to develop a minimally invasive injectable biomimetic hydrogel for cell therapies. Pluronic grafted HA was synthesized by the addition of amine end-capped pluronic F127 on carboxyl groups of HA backbone. The synthesized HA-g-Pluronic exhibited a thermoresponsive sol-gel transition over the range of 20 - 40 oC and improved mechanical properties. Mass erosion rate was compared with pluronic F127 and the result indicated that HA entanglement enhanced the stability of pluronic micelle packing. The mechanical properties and mass erosion rate were further enhanced by modifying HA with APMMA which gave free vinyl groups for photopolymerization. To encourage the cellular attachment, cyclic RGD was end-capped with PEG-acrylate and added as a crosslinker to the HA-g-Pluronic system. Cytocompatibility tests of HA-g-Plu hydrogels were conducted using bovine chondrocytes in vitro demonstrating good cell viability and proliferation. The gene expression of encapsulated cells was also tested and the re-differentiation of chondrocytes was confirmed. Finally the neocartilage formation in mouse upon the injection of cell containing HA-g-Plu was studied.
