A microbead-DNA assay based on the coffee ring effect for detection of multidrug-resistant bacteria |

The spread of bacteria and antimicrobial resistance is becoming a serious threat to public health. The recent advent of molecular techniques such as qPCR or microarrays allow specific detection of targets, but many limitations such as high costs and needs of skillful expert for their application in the clinic. Here, we propose a simple diagnostic system that can detect antimicrobial resistance in bacteria using bead based sensing of nucleic acids. Bead-oligo targeting mecA - the gene involved in methicillin resistance of Staphylococcus aureus - was prepared to detect single-strand DNA (ssDNA) products obtained by rolling circle amplification (RCA). We simply observed suppression of the coffee-ring effect by microscopy in presence of synthetic target ssDNAs or RCA products from bacteria with specificity. This molecular sensing strategy based on bead based visual detection will introduce simple and rapid platforms with high sensitivity for diagnosing infections. |
