In vitro and In vivo Evaluation of PVPA/ Chitosan Scaffold for Bone Tissue Regeneration |

In this work, simple lyophilization method was used to generate porous membrane and scaffold with polyvinylphosphonic acid (PVPA) and chitosan. PVPA contains negatively charged phosphonic groups and Chitosan is a known biocompatible polymer which contains cationic functional group. Combined PVPA/Chitosan polymer can enhance pre-osteoblast cell viability, adhesion and proliferation. Different concentrations of PVPA were prepared and combined with purified chitosan to form complexes. Lyophilized scaffolds were characterized and compared to obtain optimum condition and material property. Complex formation was confirmed by Fourier Transform Infrared Spectroscopy (FTIR) and the microstructure was analyzed via Scanning Electron Microscopy (SEM). Porosity by water absorption and pore size distribution was also obtained. Electrostatically bound PVPA desorption was determined by colorimetric determination of phosphorus content using KH2PO4 as phosphorus standard. The biodegradation with lysozyme behaviors of PVPA/chitosan scaffolds were also investigated. Cytotoxicity using MC3T3-E1 pre-osteoblast cells was evaluated by MTT assay and cell adhesion behavior was observed by confocal microscopy. In vivo study was also conducted using rats as model. |
