제목 Humoral and cellular immune response in mice injected by polyethylenimine/SARS CoV S DNA vaccine nanoparticles through intranasal route
초록 In this study, plasmid pCI-S, encoding the S gene of SARS-CoV was constructed and delivered by polyethylenimine (PEI). The expression of the S protein was confirmed in vitro. The immune responses after intranasal immunization with PEI/pCI-S nanoparticles were examined in BALB/c mice. The mice immunized with PEI/pCI-S nanoparticles produced much higher level of IgG and IgA than those treated with pCI-S alone. Spleens and lungs were isolated from the immunized mice, and then the cells were cultured. The proliferation of B220 cells and some cytokine producing cells (IFN-γ, TNFα, TNFα+ IL-2) isolated from PEI/pCI-S vaccinated mice were increased after restimulated with antigen. Co-stimulatory molecules (CD80 and CD86) and Class II MHC molecules (I-Ad) expressed by dendritic cells also showed higher levels in PEI/pCI-S vaccinated mice than pCI-S vaccinated mice. As the results, the PEI/pCI-S nanoparticles induced both humoral and cellular immune responses.
