제목 Enhancement of Immune Responses of SARS CoV S DNA Vaccine Using Biocompatible Micelle as a Gene Carrier
초록 Purpose. A novel biocompatible micelle gene delivery system was fabricated to improve its gene transfection efficiency, accordingly enhance the immune responses of SARS CoV S DNA vaccine. Materials and Methods. PEG-block-P[Glu(SPM)] (PGS) was simply synthesized. PGS/DNA micelles were characterized, and the condensation and protection ability of plasmid DNA by PGS were confirmed by agarose gel electrophoresis assay. Cytotoxicity and gene transfection efficiencies were evaluated. The expression of the S protein was confirmed in 293T cells. Results. PGS/DNA micelles showed spherical compact shapes with nanometer size. Cytotoxicity of PGS was rather lower than that of PEI 25K. And PGS/DNA shows higher gene transfection efficient both in the presence and absence of serum. PGS/pEGFP-N2 micelles also showed good gene expression in vivo. Conclusions. The results demonstrated the potential use of PGS as an effective gene carrier, and enhance the immune responses of DNA vaccines.
