Highly Specific Multiplex Detection of SNPs via Encoded Hydrogel microparticles conjugated with Universal Mismatch DNA probes |

Nucleic acid multiplex detection using encoded hydrogel microparticles is a detection technology attracting attention by its high specificity and multiple detection capabilities. In particular, single nucleotide polymorphisms (SNPs), generated by mutation of only one sequence in a nucleic acid, are well known as useful biomarkers. However, encoded hydrogel microparticles that utilize probe-target hybridization as a key mechanism are hampered by the small duplex stability differences resulting from single base-pair mismatches. We have developed encoded hydrogel microparticles with universal mismatch DNA probes for multiplex SNP detection. A widely used base analog (5-nitroindole) was used to assess the ability of the probe to detect a mutation at a given SNP site. We certified that our hydrogel microparticles with DNA probes where one base is substituted with 5-nitroindole exhibit much higher specificity for a single base-pair mismatch with minimal reduction in sensitivity. |
