제목 Harmonization of DNA codes and expressed proteins using Janus particle
초록 The Janus particle has been attracted because the particle can be conjugated with different functional groups on each hemisphere. However, the practical application of the Janus particle was extremely rare without Janus printing and some cascade catalysts. Here, we introduce practical application of Janus particle as harmonization of geno- and pheno-type of protein on single particle. The Janus platform was successively fabricated via Pt-sputtering on the amino-silane treated silica sphere. DNA including green fluorescence protein (GFP) and streptavidin-binding protein (SBP) sequence was conjugated with one side of the Janus particle via thiol-metal conjugation. And then, the other side of the particle was covered with streptavidin by EDC/NHs coupling. The specific DNA of protein was expressed in emulsion using in-vitro expression. The GFP can conjugated with the other side of Janus particle by the SBP. As the results, the Janus particle could be applied to DNA screening platform.
