Development of Acyl-Homoserine Lactone Specific Binding DNA Aptamer |

Quorum sensing is a system of cell–cell communication that allows bacteria to recognize information about cell density and regulate gene expression. Many bacteria release a variety of chemical signaling molecules called autoinducer (AI). Acyl homoserine lactone (AHL) is the most famous class of AI and is mainly made by Gram-negative bacteria. Although research on AI has been extensive, the method of detecting AI is limited and requires long time. New faster detection method are required to overcome the existing limitation. Aptamers are short single-stranded DNA or RNA that can bind to a variety of molecules with high affinity and specificity. It can maintain their structure in different condition and can be easily modified for variety of purpose. In this study, SELEX was conducted for 10 round to select AHL specific DNA aptamers. Finally, binding affinity of aptamer candidates were estimated by using SPR, and five aptamers with high affinity to AHL were selected.
