Highly sensitive, selective and multiplex sensors have become an important challenge for disease diagnosis. Here, we describe a multiplex biosensor for the detection of cancer biomarkers based on distinct plasmon response of single gold nanoparticles (AuNPs) and antibody-antigen binding activity. To demonstrate the ability to detect and quantify cancer biomarkers: a panel of biomarkers of α-fetoprotein (AFP), Carcinoembryonic antigen (CEA) and prostate specific antigen (PSA) was used as a model analytes. A multiplex biosensor was developed by site-selectively immobilizing plasmonic nanoparticles functionalized with an each monoclonal antibodies that recognize the target protein on patterned glass slide. The proposed biosensor exhibited outstanding selectivity and sensitivity. Limit of detection was determined at of 89 fM, 90 fM and 9 fM for AFP, CEA and PSA. Finally, using this sensing strategy, the platform becomes an excellent approach in both research and clinical medical fields. |
