In this article, we describe a multiplex biosensor for the detection of cancer biomarkers based on plasmon response of single gold nanoparticles (AuNPs) and antibody-antigen binding interaction. To demonstrate the ability to detect and quantify cancer biomarkers: prostate specific antigen (PSA), a-fetoprotein (AFP), Carcinoembryonic antigen (CEA), a novel and sensitive multiplex sensor was developed by immobilizing plasmonic nanoparticles functionalized with monoclonal antibodies that recognize the target protein on hydrophilic-hydrophobic patterned glass slide. The proposed multiplex biosensor exhibited outstanding sensitivity and selectivity. Using this sensing strategy, we found that multiplex ability, high sensitivity, simple, and ease of use of the detection method based on localized Surface Plasmon Resonance(LSPR) spectrum of single gold nanoparticles make it an excellent application for versatile molecular diagnostics in both research and clinical medical fields. |
