Size exclusion chromatography (SEC) developed for separation of large molecules such as proteins, polymer, peptides, nucleic acid and polysaccharide following to their molecular size. The retention factor was related to several parameters such as particle size and pore size of stationary phase and the composition of mobile phase. In this work is to determination of the retention factor of dextran (5, 25, 50, 270, 670 and 1100 kDa) and polysaccharides such as fucoidan, alginic acid and laminarin in SEC stationary phase. 1.2 is the largest retention factor in tow difference SEC column (5 kDa dextran). The smaller the pore size of the column is suitable to analyze a low molecular weight. On the other hand, large molecular weight is suitable large pore size column. The 0.03M NaOA is best buffer in fucoidan and alginic acid and no buffer is best condition in laminarin. |
