제목 Polyvinyl Alcohol Derived Preformed Hydrogel Preparation by Silk Fibroin and Dexamethasone Loading to Achieve Full-thickness Skin Wound Healing
초록 Systemic administration of corticosteroid, e.g. dexamethason (Dex) to combate massive inflammation due to skin injury, is characterized by angioedema, bradycardia, cardiac arrhythmias etc. Silk fibroin (SF), polyvinyl alcohol (PVA) and Dex were strategically blended to prepare a wound healing hydrogel. Dex was dissolved in absolute ethanol and incorporated into the composite blend. After repeated freeze-thaw exposure of the composite blend, the preformed cryogel delivery system was obtained, which can flow under moderate pressure allowing passage through a small gauge needle. The properties of PVA-SF-Dex were compared among different compositions in different experiments in which composite containing 5%PVA-5%SF possessed high drug release characteristics, complete Dex release over 14days compared to other compositions were achieved. Non-toxicity of samples were confirmed by cell viability assay. In vivo wound healing activity in rat excisional wound healing model revealed that, compared to samples without Dex, PVA-SF-Dex shows better wound healing performance, which was further supported by histological analysis.
