Although polyethyleneimine(PEI)25K is considered as a gold standard in gene delivery system, it has high cytotoxicity. In this study, Sucrose-PEI2K was synthesized by a simple method to achieve lower toxicity compared to PEI25K, maintain high transfection efficiency for gene delivery system. After sucrose was oxidized using sodium periodate, PEI2K was cross-linked with oxidized sucrose based on imine Schiff base reaction. Chemical structure of Sucrose-PEI2K was confirmed by 1H NMR and GPC. As a result of the gel retardation assay and Pico green assay, it was confirmed that the Sucrose-PEI2K condensed DNA at a weight ratio of 0.4. Cell viability of Sucrose-PEI2K was higher than that of PEI25K in MTT assay. Moreover, Sucrose-PEI2K showed higher transfection efficiency than PEI25k in vitro cell experiments. Taken together, these results support the potential of Sucrose-PEI2K for gene delivery system. |
