In the present study, we carried out solution X-ray scattering (SAXS) experiments on the Estrogen-Related Receptor Ligand Binding Domain and its complex, in order to obtain detailed information on the overall structure at the native state. The solution SAXS data were analyzed in detail, providing important information on the structure. From the SAXS profiles and determined parameters, structural models of the mERRα LBD and its complex were reconstructed, which was made inside the search volume of maximum diameter Dmax calculated from the p(r) function. Ultimately, the structural evidence may have implications in establishing relationship between the structure of mERRα LBD and its complexes and their transcriptional activation function. Furthermore, the present finding turned out the possibility that solution SAXS method could be applied to predict the spatial orientation of a structure that has not been solved by X-ray crystallography caused by the difficulty of crystal growth. |
