Application of enzyme sequence similarity network for development of potential antibiotic agents |
초록 |
Enediynes represent rare and super potent anti-cancer drugs exhibiting DNA cleavage activieis. Up to date, there are only five identified bioactive, and four bio-inactive 9 membered enediyne molecules. Stabilization of conjugated systems in the molecules are acquired by enediyne carrier proteins. A common feature in the biosynthesis of the frou bio-inactive enediynes is that there are no enediyne carrier proteins present in their biosynthgetic gene clusters. Bioinformatics analysis of multiple Streptomyces genomes were performed in order to conver the four bio-inactive enediynes into their bioactive forms by expression of heterologous carrier proteins. |
