Piceatannol and 3',4',7-trihydroxyisoflavone(3’-ODI), i.e. ortho-hydroxylated resveratrol and daidzein, have been shown to act as an inhibitor of leukemia and a potential skin cancer chemopreventive agent. Using tyrosinase from Streptomyces avermitilis (SaTyr), resveratrol and daidzein can become ortho-hydroxylated, but these products are rapidly oxidized by SaTyr again, becoming quinone compound and melanin. Herein, we did a saturation mutagenesis with residues located near around an one of copper in the active site and found a variant having improved productivity of ortho-hydroxylated compound by high through-put screening which we designed. Using homology modeling, we could make an explanation about the reason of increasing productivity of ortho-hydroxylated compounds. |
