Optimization of protein solubility for proteomic study of Streptomyces peucetius subsp. caesius ATCC 27952 |
초록 |
Streptomyces peucetius produces doxorubicin which is a clinically important chemotherapeutic agent. Recently its full genome sequence has been completely sequenced. So it is needed to study protein phenotype, regulon, stimulon, proteomic signature, etc. through protein expression profile. A two-dimensional gel electrophoresis(2-DE) reveals most of a particular expression profile. Here we show the optimal condition of the protein solubilization for 2-DE to prepare 2-D reference map. In order to maximize protein solubility of Streptomyces peucetius, the optimal combination of chaotropic agents, a reducing agent, and a detergent was developed for 2-DE. |
